Abstract

Xylella fastidiosa is a xylem-limited phytopathogen with a very slow growth grow in axenic conditions compared to other common plant pathogenic bacteria. As such, the optimization of culture media remains a major challenge for improving bacterial in vitro growth to enhance biological and molecular investigations. The development of robust analytical methods is essential for investigating its extracellular metabolism and supporting future studies on microbial interactions. In this work, 1H NMR spectroscopy was used to characterized the metabolic profiles of the supernatant fractions recovered from bacterial liquid cultures of X. fastisiosa subsp. pauca ST53. Two media with marked differences in composition and growth-supporting properties, were used: (i) XFM, a minimal medium and (ii) PD3 a richer substrate commonly used to successfully grow a broad range of Xylella strains.1 Culture supernatants were collected at five time points (0, 3, 5, 7, and 10 days), immediately centrifuged to remove bacterial cells, aliquoted, and stored at −80 °C until analysis. An operative protocol for NMR sample preparation was optimized in order to obtain reproducible and robust 1D 1H NMR spectra. Different preparation conditions and pulse sequences were evaluated. CPMG provided better baseline quality and signal resolution than NOESY, especially for XFM samples, likely due to the presence of proteinaceous components such as bovine serum albumin.2 Preliminary qualitative analysis highlighted time-dependent metabolic changes in both media, with more pronounced spectral variations in PD3, consistent with its richer composition and faster expected grow rates. Overall, this study defines a robust NMRbased workflow for monitoring X. fastidiosa culture media and provides a first metabolomic comparison of XFM and PD3, laying the groundwork for future exometabolomic and footprinting analyses aimed at identifying metabolites involved in bacterial adaptation and microbial interactions3.
